Personally, I prefer to remove the infected drive and then insert it as the secondary drive in another computer that has the latest antispyware and antivirus (Microsoft Security Essentials). Because the drive can not boot up, the virus never has a chance to run and the antivirus software on the first drive will be able to detect and remove it.
I have had 100% success removing spyware this way – in fact, I just did this last week for a family member and found five viruses, one was self generating (could grow others).
One important note when using this method – make sure that the computer does not boot from the infected drive or you could end up infecting the good system.
